(tylko po angielsku) The main objective of this module is to acquaint students with the usage of immunostaining method in detection of the epigenetic chromatin modifications in plants. The method is based on the in situ aplication of specific antibodies against methylated histones and DNA. The results of this method are acquired with confocal microscopy and can be studied in any type of plant cells or tissues. IMMUNOSTAINING method is commonly used for many plant species, particularly in epigenetic chromatin modifications studies. Commercially available antibodies are invaluable tools for localizing different types of histone and DNA modifications in situ (Fuchs et al., 2006). Most data on global distribution of histone methylation marks in plants are available for Arabidopsis thaliana. Immunostaining with antibodies that discriminate between mono-, di- and trimethylation of specific lysines. H3K4me1,2,3 and H3K36me1,2,3, together with H3K9me3, H3K27me3 and H4K20me2,3 were identified as typical marks for euchromatin, by contrast, the heterochromatic chromocentres of Arabidopsis were found to be enriched in H3K9me1,2, H3K27me1,2 and H4K20me1 (Soppe et al., 2002, Jasencakova et al., 2003, Fuchs et al., 2006). Some data are also available for chromosomal localization of epigenetic histone modifications (Shi and Dawe, 2006), (Houben et al., 2003). |
